Je trojni agonist receptorjev za GLP-1 (glukagonu podoben peptid-1), GIP (\u017eelod\u010dni inhibitorni polipeptid, znan tudi kot glukozno odvisni insulinotropni polipeptid) in GCGR (glukagonski receptor). Retartrutid je peptid s 39 aminokislinami, ki je odporen na cepitev z DPP4, encimom, ki cepi GLP-1 in GIP.
